"Noooo" is just a way of writing that they're dragging out the vowel sound of "No!". The emotion they're conveying is extremely dependent on context. Could be anger, disappointment, despair, etc. It all depends on the person and what they're responding to.

Sep 29, 2021 · He mostly breathes and indimidates with occasional hints of Anakin's dark humor. That being said, I believe that if Vader is silent when in murderous mood which is most of the time then his input has a lot of weight. I don't like this whole "noooo" trope in cinema, it's corny and I get it why people find Vader's two NOOOO's cringy.

Vader's voice is not simply an amplified version on what he says, its more like a Stephen Hawking system whereby a computer recognizes what Vader is trying to say (His voice is probably extremely damaged and unrecognizable), then basically says it for him.
