In the U.S. state of Kansas, US-40 is a main east–west highway that runs from the Colorado border east to the Missouri border. The highway is concurrent with Interstate 70 (I-70) from Oakley to Topeka, and again through Kansas City. US 40 enters Kansas near the unincorporated community of Weskan.

US 40 passes through or by major cities including Denver, Kansas City, St. Louis, Indianapolis, Columbus, Baltimore, and Wilmington. Three former and four current state capitals lie along the route.

The only access from Alt. US 40 back to US 40 (which multiplexes with I-70 most of the way across Kansas, basically from Junction City eastward) is by crossing the highway here. US 40 and K-4 WB on the east side of Topeka, where both enter I-70 to pass through the city at Exit 366.

The original plan for US 40 through Kansas called for it to be routed on the Golden Belt Highway from Kansas City, through Lawrence and Topeka, to Manhattan, then west on the Midland Trail via Colby and Goodland before hitting the Colorado line at Kanorado.

This pamphlet by the U.S. 40 Highway Association promotes U.S. Route 40 as the trans-continental highway from Atlantic City, New Jersey, to San Francisco, California. At the pamphlet's creation, the highway had not yet been completed and reached to eight miles west of Topeka, Kansas.
