Chloroquine inhibits autophagic flux by decreasing ... - PubMed
Dec 20, 2019 · We show that CQ mainly inhibits autophagy by impairing autophagosome fusion with lysosomes rather than by affecting the acidity and/or degradative activity of this organelle. Furthermore, CQ induces an autophagy-independent severe disorganization of the Golgi and endo-lysosomal systems, which might contribute to the fusion impairment.
Chloroquine-Induced Accumulation of Autophagosomes and …
Chloroquine (CQ) is an antimalarial drug known to inhibit autophagy flux by impairing autophagosome–lysosome fusion. We hypothesized that autophagy flux altered by CQ has a considerable influence on the lipid composition of endothelial cells.
Chloroquine treatment induces secretion of autophagy-related
Feb 28, 2022 · We showed that lysosomal and autophagy inhibition by CQ altered the secretome, and induced the release of Atg8 orthologs and autophagy receptors. Atg8-family proteins, in particular, were secreted inside small extracellular vesicles (sEVs) …

Jul 20, 2018 · In this study, we focus on how CQ inhibits autophagy and directly compare its effects to those of BafA 1. We show that CQ mainly inhibits autophagy by impairing autophagosome fusion with lysosomes rather than by affecting the acidity and/or degradative activity of this organelle.
Inhibition of autophagy by chloroquine prevents resistance to …
Jun 27, 2022 · We, therefore, investigated if two PI3K/AKT inhibitors, ipatasertib and taselisib, could induce autophagy in breast cancer models, and whether chloroquine (CQ), a well known autophagy inhibitor, could potentiate ipatasertib and taselisib anti-cancer effect in combination with conventional chemotherapy.
Regulation of autophagy and chloroquine sensitivity by …
Chloroquine (CQ) is an antimalarial drug and late-stage inhibitor of autophagy currently FDA-approved for use in the treatment of rheumatoid arthritis and other autoimmune diseases.
The clinical value of using chloroquine or hydroxychloroquine as ...
CQ and HCQ can inhibit autophagy by blocking the fusion of autophagosomes with lysosomes. In addition, CQ also has some autophagy-independent anticancer effects, including sensitizing cancer cells to chemotherapy.
Autophagy in tumor immune escape and immunotherapy
Mar 19, 2025 · Research also indicated that CTLA-4 in Tregs can be degraded through the autophagy-lysosome pathway, with autophagy inhibitors like CQ identified as inhibitors of CTLA-4 degradation . In conclusion, the expression levels of CTLA-4 are closely related to autophagy, which have an impact on the TIME and immune responses. Autophagy in macrophages
